Probang Cups
Collection of O/P Fluid
Reconstitution of antibiotics

O/P fluid is collected using a probang cup. This is a cup-shaped
piece of brass attached to a wire handle.
Prior to sampling with a probang cup, 2 ml of 0.08 M phosphate buffer
(Eagle's MEM or Earle's LYH tissue culture medium with 0.04 M HEPES buffer
is also satisfactory) containing 0.01% bovine serum albumin, phenol red
(0.002%) and antibiotics adjusted to pH 7.2, should be added to a small
bottle or similar container for each of the animals to be sampled. Each
bottle should be identified with a waterproof label.
Collection of O/P fluid
• restrain the animal and open the mouth.
• pass the probang cup over the root of the tongue.
• place one hand over the pharyngeal area and continue to introduce
the probang cup until it can be palpated in the upper part of the oesophagus.
• pass the probang cup backwards and forwards several [5-10] times,
each time pushing far enough forward to push the cup into the cranial
oesophagus as evidenced by slight resistance to forward movement.
• gently withdraw the probang cup, keeping it upright to ensure that
the O/P fluid remains inside.
• visually inspect the material.
