Appendix 4

Transport Medium

Medium for collection of epithelium and vesicular fluid

0.04 M phosphate buffer

Add 3.05 g Na₂HPO₄.2H₂O

0.39 g KH₂PO₄
to 500 ml of sterile distilled water.

Add 1 ml of 1% phenol red.
Add antibiotics
Adjust pH to 7.2-7.6 with HCl.

Equal volume of glycerol

Medium for collection of probang samples

0.08 M phosphate buffer

Add 6.11 g Na₂HPO₄.2H₂O

0.78 g KH₂PO₄
to 500 ml of sterile distilled water.

Add 1 ml of 1% phenol red.
Add antibiotics
Adjust pH to 7.2-7.4 with HCl.

Reconstitution of antibiotics

Penicillin phial of 50 0000 units, add 2.5 ml sterile distilled water
Mycostatin phial of 50 0000 units, add 10 ml sterile distilled water
Neomycin phial of 50 0000 units, add 10 ml sterile distilled water
Polymyxin phial of 35 0000 units, add 7 ml sterile distilled water

To each 500 ml of 0.08 M or 0.04 M phosphate buffer add the following amounts of reconstituted antibiotics:
Penicillin 2.5 ml (final concentration 1000 units/ml)
Mycostatin 1.0 ml (final concentration 100 units/ml)
Neomycin 1.0 ml (final concentration 50 units/ml)
Polymyxin 0.5 ml (final concentration 100 units/ml)
